      First Human Trial of an Oral Native Testosterone: Physiological Levels in Fed and Fasted States (2020)This phase 1b, single-center, randomized crossover trial evaluated the pharmacokinetics of DITEST, an oral lipid-based native testosterone formulation, compared to oral testosterone undecanoate (TU).Methods: Single doses of DITEST (120 mg or 200 mg) or TU (80 mg) were administered after a high-fat meal (Part 1), followed by 200 mg DITEST in fed vs. fasted states (Part 2). Blood samples were collected over 10 hours for testosterone and DHT levels (baseline-adjusted).
      Participants: 25 adult men with hypogonadism (one withdrew after one period, included in safety only); aged 18–80, BMI 18–35 kg/m², baseline testosterone <8 nmol/L.
      Results: DITEST showed dose-proportional absorption, with 200 mg achieving Cmax of 30.4 nmol/L and AUC0-10h of 88.6 h·nmol/L—equivalent to TU but with lower DHT (Cmax 1.31 nmol/L vs. 6.7 nmol/L for TU). Absorption was food-independent (fed/fasted Cmax ratio ~0.87). One serious adverse event (urinary retention) occurred with TU; overall safety was comparable, with no new concerns for DITEST.
      Conclusions: Oral native testosterone (DITEST) delivers physiological levels irrespective of food, with reduced DHT elevation risk compared to TU, supporting further development for hypogonadism.

      ngredients in DITEST (DNL-0300) CapsulesDITEST (now referred to as DNL-0300) is an oral lipidic formulation of native (unesterified) testosterone, developed as an immediate-release soft gelatin capsule containing 40 mg of testosterone per capsule. The composition is designed for food-independent absorption, using a lipid solution with co-solvents for solubilization. Below is the full breakdown of ingredients based on the formulation details from clinical studies.Ingredient

      The formulation is stable for 2 years at ambient temperature (25°C) and is packaged in aluminum foil blister packs. This matches descriptions from earlier patents and trials, confirming the excipients (sesame oil, ethanol, benzyl alcohol, and propylene glycol monolaurate) support targeted hepatic delivery with minimal systemic DHT elevation.

        I was looking at this idea awhile ago. In theory if you could figure out the solvent part of it without killing yourself, then mix the dissolved test in oil, it could bypass the liver and conversion to DHT and other useless metabolites.

        I think if you want to go supraphysiological though you'd need some kind of 5-ar inhibitor like dutasteride. I'm also not sure if test dissolved in ethanol is going to mix well with oil without that propylene glycol, and then there's the question of whether you can swallow the ethanol without burning a hole in your esophagus.

        Might just have to wait for these Big Pharma capsules, which will require a prescription and cost a trillion dollars no doubt.

          the idea has existed for a long, long time,

          Andriol capsules are Testosterone Undecanoate in castor oil.

          Lance armstrong used to mix Testosterone Undecanoate into olive and oil drink it.

          Peat even mentioned using testosterone base in Vitamin E

            @wester130 Yeah but I think if you don't dissolve the powder it's not useful. And I've tried oil and vitamin e. They don't dissolve test powder.

              @Crypt-Keeper

              a few drops of everclear would have made it dissolve

              28ml vitamin e

              2ml ethanol like everclear

                @Crypt-Keeper lots of hormone powders including various test esters dissolve fine in just plain olive oil . you melt the powder first and then add the oil.

                your post is overall kinda weird talking about people potentially killing themselves with alcohol and trillion dollar capsules. please get a grip
